What is the protocol for surface-treatment of coverslips?
PLL procedure:
You can watch a video of this procedure at : https://www.youtube.com/watch?v=mN2XfIiEmiQ
Materials:
Poly-lysine 0.01% in H2O (Sigma-P4832, ready-made; Sigma-P2658, powder)
Milli-Q H2O
Cleaned high precision coverslips (Thorlabs-CG15KH)
Pressured air, to dry coverslips
Procedure:
Place two pre-cleaned coverslips over a lint-free and clean surface.
Label the upper left corner of each coverslip (to later identify the coated surface).
Place a drop of 0.01%poly-lysine in the center of one coverslip.
Flip the second coverslip and place over the drop perpendicular to the 1stcoverslip.
Incubate for 30 sec and then separate the two coverslips.
Dip into flask with MQ-H2O to get rid of excess poly-lysine.
Wash twice with MQ-H2Ostream directed at the treated surface.
Dry coverslip with clean pressured air.
Place a reusable gasket on top of the treated surface and use immediately or store in a clean container and use within a week.
