What factors can affect the focus?
Some of the factors that affect the focus and/or lead to focus drift are:
Adding cold sample to RT buffer
Oil or buffer getting under the coverslip
Too much or too little oil
The autofocus camera settings
If you find the focus is not behaving at all its likely that:
Oil has gotten under the coverslip and sample stage
The gasket has leaked
The autofocus camera exposure time is not optimised
