How do you know if the auto-focus needs calibrating?
You only need to repeat the auto-focus calibration if the AcquireMP settings have been reset.
Before you run the autofocus calibration, add Milli-Q water or clean buffer and move the sample into focus so that the ring is visible. Make sure the ring is well centred. If not adjust the offsets in the Auto-Focus Camera Settings.
