Sample mass changing between wells
Troubleshooting question
I've measured the same sample across different wells/gaskets using the same mass calibration and the mass sometimes changes by 20-30 kDa when I change move. Not sure if this an issue with the machine, the focus, coverslip or something else. Has anyone seen and resolved this before?

Quick question - was the system on for more than an hour before you made a measurement? An hour is about the time need for all internal temperatures and laser powers to be optimal. It might have been not quite in focus.
Alternatively, it might have been the buffer (was there anything different to salt in this?) and was the calibration run in the same buffer?
Additionally, did you run more than 2 repeats? It's always worth trying a few to make sure. Finally, it may be worth trying a full "Recalibrate Image" (AMP, Settings > Acquisition) to make sure the settings are right.
