Inverted signal
Hi all,
Our instrument seems to have a recurrent problem where suddenly in the middle of a series of experiments the signal intensities suddenly become inverted, i.e. positive in the center. We have seen this for different samples and different users on different days, so we are pretty sure it is a problem with the instrument rather than the samples. The problem is usually fixed if you close down the instrument and try again the next day.
Does anyone else experience this and is there a known solution?

It sounds like you are focusing on the wrong contrast maximum. There are two maxima that lie close together and the one with the objective closer to the sample (at least on our coverslips) is always the correct one and the correct one is about 30% higher in contrast than the wrong maximum.
